3.5" desktop drives normally require a 12 volt supply and some draw current up to .65A.
So, from your later postings it seems that it is a 12V desktop with a 12V supply. So what do you mean by disconnecting the 5V supply? It doesn't draw on this power. However some USB connections use the PS supply pin out to create a handshake between PC and device when connecting. This is normally between PC and DAC but I am not dismissing that it may occur with a drive too. If so the PC 5V supply ( not the external supply as this cannot create the handshake)can be disconnected after the handshake has taken place as the supply is then not used.
So you are not really connecting and disconnecting a 5V power supply but may be preventing the handshake and that is why it isn't working. However there won't be any sonic benefits from disconnecting as the 5V PS isn't actually supplying the drive.
