Yes, it should hold

The subwoofer boxes I made for my cars are 3/4" MDF. Nothing but butt joints, carpenter's glue, and a sh*tload of screws. These boxes are extremely strong. Needless to say, I wouldn't bother making joints - just butt the pieces together, glue and screw. Attempting to make joints could potentially make the design weaker.

You may want to drill small pilot holes for the screws. It will help prevent you from splitting the ends of the MDF.

If you still have strength concerns, you could use 3/4" plywood for the sides and bottom. As Tubes n' Hornz pointed out, depending on how deep you end up making the box, additional bracing could be needed. I would think angle brackets around the joints would do the job, as would the metal bands mentioned.
