Not always, as I note at the bottom. As to "break in", where no solder is involved, I have yet to perceive a major difference between similar cables (one set new and the other in use for several weeks). The contact area is what seems to "mature" over a short span of time. This is more obvious to me because I bi-amp; it is the tweeter clarity that gets (peceivably) affected. Most people would hardly notice it. BTW, it is more obvious with interconnects.
However, where solder is used, I have learned to give them time. The wire itself seems to be inert but something does appear to happen with use over time at solder joints. Things smooth out at the top end. In fact, just reheating the solder can reset the cycle.
There are good cables and bad cables. And, there are cables that simply sound different than others. There is no reason for ANY of them to cost thousands of dollars.
However, I have spent hundreds making my own. Mostly, it happened because of the amount of Magwire packages that I had to buy (for the specific guage mix required). Not a single regret...well, the final incarnation, as ribbons, ain't easy to conceal but that's another matter.

I've had a couple of instances where things got worse with time. One of these happened to a friend and myself at about the same time. It was with 12GA (electrical wiring) wire. A day after he started to use it, he called me and insisted that I had to try it. He brought me some. I did not like its sound from the beginning but agreed to give it a week. By the end of the week we both agreed that it sounded worse still. Totally different systems (his costing $25k plus, mine a pair of MMGs and cheap gear).
The other was one of the various "Cat 5" incarnations. After all the work putting it together, I really wanted to like it. This one included soldering. Two weeks later I could not stand it. My take, the soldering cured to reveal more of the "bad".
