As Stephen says, you can solder and unsolder connections many times, so long as you're careful.
Two suggestions: If the connections are large tabs, the solder wick may take longer to soak up the solder, allowing more heat to poss. damage the tab / VC leads. Use some 000 steel wool (small ball of it) w/ some rosin flux dabbed into it. Hold it w/ tweezers against the joint then quickly heat. The steel wool will soak up lots more solder than the wick, thus, faster desoldering and less heat damage.
Two, if you're going to be taking the speakers apart many many times, why not just use slip-on connectors until you get the results you're looking for (ie; damping, etc) then snip off the connectors and solder the wires on permanently.
