There is nothing inherent in the design of a typical speaker cable that would render it directional. However, I do believe that it will develop directionality as a result of use and break in, but this directionality is reversible.Some claim that the direction the wire went through its die during its manufacture will determine the directionality of the cable, but I have not found this very convincing.
